Mitochondrial DNA Maintenance Disorders: Impact of Impaired Deoxynucleoside Triphosphates Metabolism

چکیده
The maintenance of mitochondrial DNA (mtDNA) is dependent upon several nuclear gene-encoded proteins including enzymes forming the replisome needed to synthesise mtDNA. These need be present in balanced quantities function properly. In addition, mtDNA synthesis requires a supply nucleotides that achieved by nucleoside recycling inside mitochondria, and nucleotide import from cytosol. Mitochondrial defects are group diseases caused pathogenic variants genes involved maintenance, result impaired replication. Pathogenic gene identified date include encode replication machinery (such as POLG), help maintain pool TK2), fusion. Here, presentation provided Ramon Martí, Research Group on Neuromuscular Diseases, Vall d’Hebron Institut de Recerca, Barcelona, Spain, CIBERER, Madrid, summarised. A leading expert pathology, Martí presented at Euromit 2023 International Conference Disease, which took place Bologna, Italy, May 2023.

Rapid Hydrolysis of Deoxynucleoside Triphosphates Accompanies DNA
Bacteriophage T4 gene 44/62 and 45 accessory proteins stimulate DNA synthesis by T4 DNA polymerase. Using a @X174 DNA template primed with a strand of a restriction fragment, these accessory proteins also greatly increase the hydrolysis of dNTP to dNMP which accompanies DNA synthesis by the polymerase. The rate of formation of a deoxynucleoside monophosphate is up to 10 times the rate of its in...
متن کاملQuantitation of cellular deoxynucleoside triphosphates
Eukaryotic cells contain a delicate balance of minute amounts of the four deoxyribonucleoside triphosphates (dNTPs), sufficient only for a few minutes of DNA replication. Both a deficiency and a surplus of a single dNTP may result in increased mutation rates, faulty DNA repair or mitochondrial DNA depletion. dNTPs are usually quantified by an enzymatic assay in which incorporation of radioactiv...
متن کامل5-Hydroxypyrimidine deoxynucleoside triphosphates are more efficiently incorporated into DNA by exonuclease-free Klenow fragment than 8-oxopurine deoxynucleoside triphosphates.
Recent studies with 8-oxodeoxyguanosine triphosphate (8-oxodGTP) have suggested that incorporation of oxidized nucleotides from the precursor pool into DNA may have deleterious effects. Here we show that 5-hydroxydeoxycytosine triphosphate (5-OHdCTP) and 5-hydroxydeoxyuridine triphosphate (5-OHdUTP) are more efficient substrates than 8-oxodGTP for Escherichia coli DNA polymerase I Klenow fragme...
متن کاملAutosomal disorders of mitochondrial DNA maintenance.
Mitochondrial DNA (mtDNA) is maternally inherited. After birth, secondary mtDNA defects can arise. MtDNA depletion is a reduction in the amount of mtDNA in particular tissues. Multiple deletions of mtDNA accumulate as somatic mutations in mainly postmitotic tissues. These disorders of mtDNA maintenance frequently show Mendelian inheritance. Positional cloning has identified several genes involv...

The use of L(-)SddC [beta-L-2',3'-dideoxy-3'-thiacytidine (lamivudine, 3TC)] for the treatment of Herpes B virus (HBV) infection is hindered by the emergence of drug-resistance associated with the L526M, L550V, and L526M/M550V mutations of the viral DNA polymerase (DP). The interactions of the anti-HBV compounds 2',3'-dideoxy-2',3'-didehydro-beta-L(-)-5-fluorode-oxycytidine and 2'-fluoro-5-meth...
